Occasionally, small volumes of HPN ELISA kit vials may become trapped in the product vial seal during shipping and storage. If necessary, briefly centrifuge the vial in a benchtop centrifuge to dislodge any liquid on the lid of the container. The concentration gradients of the kit standards or positive controls provide a theoretical detection range of the kit in biological research samples containing PNH.

The analytical biochemical ELISA technique of the MBS287067 kit is based on HPN antibody-HPN antigen interactions (immunoadsorption) and an HRP colorimetric detection system to detect HPN antigen targets in samples. The ELISA Kit is designed to detect native, non-recombinant PNH. Appropriate sample types may include secretions and undiluted body fluids and/or tissue homogenates. Quality control trials evaluating reproducibility identified the intra-assay CV (%) and inter-assay CV (%).

This assay uses a two-site sandwich ELISA to quantify PNH in samples. A microplate has been pre-coated with an antibody specific for PNH. Standards and samples are pipetted into the wells and any PNH present binds to the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PNH is added to the wells. After washing, streptavidin-conjugated horseradish peroxidase (HRP) is added to the wells. After washing to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and the color develops in proportion to the amount of PNH bound in the initial step. The color development is stopped and the intensity of the color is measured.
